چکیده

Small extracellular vesicles (EVs) are important mediators of intercellular signaling that carry biologically active protein payloads relevant in wound healing. However, whether EVs analyzed from fluid or other biological fluids, heterogenous, reflecting part the release different cell types. To address central question EV heterogeneity parallel with unmet need forin vivo sources low platelet contamination, we have developed a macroporous scaffold for subcutaneous implantation and subsequent collection is applicable study activity any defined mouse model. Using polyvinylalcohol (PVA) sponges as scaffold, first show rapid infiltration immune cells hematopoietic origin accompanied substantial heterogenous population EVs. Second, how single vesicle flow cytometry (vFC) addresses challenge by quantifying expression surface markers map to specific subpopulations cellular origin. Third, vitro two-dimensional culture, although common, introduces significant bias addressed PVA vivomodel. Together these studies, define novel model establishing biochemical basis biology

Association of Extracellular Membrane Vesicles with Cutaneous Wound Healing

Extracellular vesicles (EVs) are membrane-enclosed vesicles that are released into the extracellular environment by various cell types, which can be classified as apoptotic bodies, microvesicles and exosomes. EVs have been shown to carry DNA, small RNAs, proteins and membrane lipids which are derived from the parental cells. Recently, several studies have demonstrated that EVs can regulate many...

Rabbit calvarial wound healing by means of seeded Caprotite scaffolds.

Autologous bone is the most successful bone-grafting material; however, limited supply and donor site morbidity are problematic. Synthetic bone substitutes are effective, but healing is slow and unpredictable. Osseous wound healing may be enhanced if bone substitutes are combined with autologous bone marrow cells. To test this hypothesis, we created 40 calvarial defects in 20 12-week-old New Ze...

Extracellular Vesicles as Neprilysin Delivery Syemory Improvement of Alzheimer’s Disease

Alzheimer’s disease (AD) is a neurodegenerative brain disorder which has no effective treatment yet due to the blood barrier in the brain that limits the drugs with the potential of disease improvement. Extracellular vesicles (EVs) are biocompatible nanoparticles with a lipid membrane. These vesicles are secreted from various cells such as mesenchymal stem cells (MSCs) and can pass through biol...
